Product Description & Details
Overview & Usage Guidance
Product Introduction: AJAPRIDE-PM 1
AJAPRIDE-PM 1 is a widely prescribed triple-combination antidiabetic medication manufactured by Ajantha. Composed of 1mg of Glimepiride (a sulfonylurea), 500mg of Metformin Hydrochloride Sustained-Release (SR), and 15mg of Pioglitazone Hydrochloride (a thiazolidinedione), it is indicated for glycemic control in adult patients diagnosed with Type 2 Diabetes Mellitus when double combination therapy is insufficient.
Primary Indications
- Management of Type 2 Diabetes Mellitus in adults for optimal glycemic control.
- Improving insulin secretion, reducing insulin resistance, and restricting hepatic glucose production.
- Reducing the long-term risk of diabetic microvascular and macrovascular complications.
Clinical Benefits
- Triple Pathway Control: Glimepiride stimulates insulin release, Metformin reduces hepatic glucose, and Pioglitazone enhances insulin sensitivity.
- HbA1c Optimization: Assists in achieving and maintaining target HbA1c (glycosylated hemoglobin) levels.
- Sustained-Release Metformin: Smooth, gradual drug release minimizes gastrointestinal side effects.
Possible Side Effects
Common side effects may include:
- Gastrointestinal issues (nausea, metallic taste, abdominal bloating, transient diarrhea).
- Weight gain or peripheral edema (fluid retention) due to Pioglitazone.
- Hypoglycemia (low blood sugar), especially when co-administered with sulfonylureas or insulin.
How to Take AJAPRIDE-PM 1
- Take the tablet daily, with or immediately after your morning meal to minimize stomach upset and prevent hypoglycemia.
- Swallow the tablet whole with a glass of water. Do not crush, chew, or split.
- Do not stop taking the drug abruptly without consulting your endocrinologist.
Mechanism of Action
AJAPRIDE-PM 1 works through three distinct mechanisms:
- Glimepiride: Binds to ATP-sensitive potassium channels on pancreatic beta-cells, causing depolarization and opening of calcium channels, triggering the release of insulin.
- Metformin: Decreases hepatic glucose production, decreases intestinal absorption of glucose, and improves insulin sensitivity.
- Pioglitazone: Activates peroxisome proliferator-activated receptor-gamma (PPAR-gamma), regulating genes involved in glucose control and lipogenesis, increasing peripheral insulin sensitivity.
Safety Warnings
- Lactic Acidosis: Metformin carries a rare but serious risk of lactic acidosis. Avoid in severe renal impairment.
- Fluid Retention: Pioglitazone can cause fluid retention, which may lead to or exacerbate congestive heart failure. Contraindicated in NYHA Class III/IV heart failure.
- Pregnancy & Nursing: Strictly contraindicated. Avoid use.
Patient Advice
- Always carry a rapid-acting source of sugar (like glucose powder or fruit juice) in case of hypoglycemia.
- Monitor your weight and check for signs of swelling in your lower limbs. Report sudden weight gain immediately.
- Ensure regular monitoring of liver enzymes, kidney function, and HbA1c levels.
